Search for functional ingredients affecting glutathione redox system and its application to ferroptosis regulation

Research Outline

Cells are constantly exposed to reactive oxygen species (ROS) generated by endogenous and exogenous stimuli. ROS are eliminated by the coordinated action of various antioxidants and antioxidant enzymes but excessive ROS causes oxidative stress and involved in various diseases. Glutathione (GSH), a major antioxidant in the cell, plays a pivotal role in ROS elimination, and GSH depletion cause iron-dependent cell death called as ferroptosis. Ferroptosis is thought to be involved in cancer and neurodegenerative diseases, and research aimed at its therapeutic application is carried out worldwide.
I am addressing the research of supply pathway of cysteine, which is rate-limiting amino acid for GSH synthesis, and GSH maintenance in the cell. The purpose in this study is to clarify the relationship between ferroptosis and disease such as cancer and neurodegenerative diseases, and search for food-derived functional ingredients that regulate ferroptosis, and investigate the effect for the maintenance and promotion of health of the ferroptosis regulate-functional ingredients. We will promote international joint research with the German research group that leads this research field, and while receiving advice and support, we will address to elucidate the mechanism of ferroptosis by searching for food-derived functional ingredients that affect the GSH redox system.
